cGMP and PKGI are required for vascular BMP signaling

چکیده
Background Maintenance of vascular homeostasis depends on phenotypic switching of vascular smooth muscle cells (VSMCs) during development, vascular injury repair, and disease. In healthy blood vessels, VSMCs exhibit a differentiated, ‘contractile’ phenotype, but in diseased vascular tissue or after vascular injury, they de-differentiate into a ‘synthetic’ state, characterized by decreased smooth muscle (SM)-specific gene expression and increased proliferation and motility [1].

Proteolytic processing of cGMP-dependent protein kinase I mediates nuclear cGMP signaling in vascular smooth muscle cells.
Cyclic GMP modulates gene expression in vascular smooth muscle cells (SMCs) in part by stimulating cGMP-dependent protein kinase I (PKGI) and the phosphorylation of transcription factors. In some cells, cGMP increases nuclear translocation of PKGI and PKGI-dependent phosphorylation of transcription regulators; however, these observations have been variable, and the mechanisms mediating nuclear ...

Nitric oxide and cGMP modulate vascular smooth muscle cell (SMC) phenotype by regulating cell differentiation and proliferation. Recent studies suggest that cGMP-dependent protein kinase I (PKGI) cleavage and the nuclear translocation of a constitutively active kinase fragment, PKGIγ, are required for nuclear cGMP signaling in SMC. However, the mechanisms that control PKGI proteolysis are unkno...

cGMP-dependent protein kinase I (PKGI) is an important effector of cGMP signaling that regulates vascular smooth muscle cell (SMC) phenotype and proliferation. PKGI has been detected in the perinuclear region of cells, and recent data indicate that proprotein convertases (PCs) typically resident in the Golgi apparatus (GA) can stimulate PKGI proteolysis and generate a kinase fragment that local...

The cyclic GMP (cGMP) signaling pathway is a ubiquitous second messenger system that regulates diverse cellular functions. The cGMP-dependent protein kinase G (PKG) is a primary direct cGMP effector in multiple cell types. cGMPPKG signaling plays critical roles throughout the cardiovascular system, although the best understood function of PKG is as an inducer of vascular smooth muscle relaxatio...

Nitric oxide signaling has an important role in regulating pulmonary development and function. Expression of soluble guanylate cyclase (sGC) and cGMP-dependent protein kinase I (PKGI), both critical mediators of nitric oxide (NO) signaling, is diminished in the injured newborn lung through unknown mechanisms. Recent studies suggest that excessive transforming growth factor-beta (TGF-beta) activ...
